on a 1975 XS650 the oil filter is held in by a large nut with a slotted screw, what does this adjuster screw do and how do you set it right?

That is the pressure adjustment for the oil filter by pass, which is simply a spring loaded ball bearing under the screw.

The valve should open with 14 PSI of pressure difference between the inlet to the filter housing and the oil galley.

The general consensus is that if the screw is set flush with the top of the bolt, that is correct. That is not fully tight as the screw can be turned in more and set below being flush with the bolt top. Some members had checked theirs when the bikes were new and found them set flush so that's pretty much how we do it since there really is no factory documentation on this.
